What is a Circadian Rhythm and Biological Clock ?
Circadian rhythms are the physical, mental or behavioural changes experienced by an organism over a cycle of 24 hours. It influences various important functions in the human body like:
- Sleep patterns
- Appetite and digestion
- Hormone release
- Temperature
The system involved in regulating any organism's sense of time controlling the circadian rhythm is called a biological clock. It acts as an essential timekeeper which controls daily rhythm for humans. Melatonin production process after onset of darkness
Melatonin synthesis as well as secretion is enhanced by darkness and inhibited by light. Some of the scientific information regarding melatonin secretion is as follows:
• Luminous information is transmitted mainly from the retina to the pineal gland through the suprachiasmatic nucleus (SCN) of the hypothalamus.
• In human melatonin secretion starts soon after onset of darkness reaching a peak at midnight gradually decreasing during the 2nd half of the night.
• Almost 80% of the melatonin is synthesized at night where serum varying between 80 as well as 120 pg/ml. During the morning the serum concentration are low like 10-20 pg/ml (Tordjman et al.).
How is Melatonin connected to our Circadian Rhythm?
Melatonin regulates the circadian rhythms like the sleep wake rhythm, neuroendocrine rhythms or the body temperature cycles through its action on the MT1 as well as MT2 receptors. However, a change in the circadian rhythm automatically affect melatonin production. If people are awake all night, then the constant exposure to blue light hampers melatonin production.

- According to 2025 systematic review published in the Journal of Public Health among 68000 individuals, almost 25.7% Indians suffer from insomnia while almost 37.4% are affected by obstructed sleep apnoea (OSA). Another 10.6% of the population suffers from restless leg syndrome a condition which disrupts sleep cycles. This is becoming a growing threat in today’s world of aspiring business and corporate hustles.
- Due to post- Covid transformation, here has been a huge change in the current working culture especially where night shifts have become very common. According to the Economic Times, India has seen almost a 15% increase in people adopting to the night shifts. Moreover a rapid growth in Indian economy has lead to a rise in the demand for night shift workers in vivid industries.
Effect of night shifts on circadian rhythm
One such study conducted among shift workers force abrupt changes in the timing of sleep as well as light dark exposure. These disturbances affect the endogenous circadian system resulting in a misalignment with the environment. Evidence based research further shows that disruption of the circadian rhythm of the night shift workers causes internal desynchronization between various levels of the circadian system. All circadian rhythms and sleep wake disturbances pinpoints the disturbing all encompassing impact of the night shift work which could contribute to the increased risk of various medical conditions (Boivin et al., 2021).
Effect of doom-scrolling on circadian rhythm
Even not in night shift today’s youth prefer to stay all night and sleep during the day. In general, after several days of staying awake at night, most metabolites shift by several hours while working at night thus leading to the misalignment with the circadian rhythm.

Studies shows that aligning work schedule against a person’s biological clock is detrimental to overall employee well being, creativity and productivity. Since circadian rhythms control major processes like sleep, cognition and metabolism any disturbance is now considered as a major factor affecting worker productivity influencing the overall economic performance.
How are Melatonin and our circadian rhythm connected
A circadian rhythm helps in the regulation of sleep, energy levels and alertness. Connecting with other countries timelines cause circadian misalignment which is one of the most consistent and alarming biological factors impairing employee performance.
It is directly connected to sleep deprivation since staying awake outside one’s biological clock definitely impacts sleep quality then sleep deprivation. Studies further confirm that sleep deprivation results from work schedules which conflict with the natural circadian rhythms. This slowly disrupts autonomic regulation increasing activation of the HPA or hypothalamic- pituitary-adrenal axis.
This dysregulation leads to a cascade of harmful effects in our body which can be summarised as follows:
• Intensified chronic fatigue
• Impaired key cognitive function like decision making, sustained attention, problem solving, working memory
• Decline in cognitive memory
• Rise in cortisol levels
• Creation of a self-reinforcing stress cycle
• Insufficient sleep recovery
• Reduction in workplace efficiency
Thus individuals with irregular sleep patterns exhibit higher cortisol activity as well as reduced melatonin production weakening emotional regulation thus increasing susceptibility to psychological distress. Another term commonly connected with this situation is the social jet lag which is the misalignment between biological time and socially imposed schedules. This increases psychological discomfort contributing to cumulative sleep debt (Poormoos, Amerzadeh, Alizadeh, Kalhoret, 2024).
Role of Magnesium in fixing the circadian rhythm
• Magnesium supports melatonin synthesis by enhancing the activity of N acetyltransferase whose deficiency lowers melatonin levels in animal studies
• Higher magnesium intake is proven to be connected with better sleep quality as well as reduced short sleep
• Mg is essential to insulin signalling and glucose metabolism closely linked to the circadian clock thus providing a plausible pathway to improved metabolic health.
